Diversity chemical and genomic of Lychnophora pinaster MART., an important medicinal specie of Brazilian Savannah

Abstract

Lychnophora pinaster (arnica-mineira) is an aromatic and medicinal species, endemic to the State of Minas Gerais and at risk of extinction; it is used by the local population in the form of alcoholic preparations of branches, leaves and inflorescences, in the treatment of bruises, sprains and hematomas. Scientific studies, especially with organic extracts of aerial parts, have shown trypanocidal, antimicrobial and anti-inflammatory activity, corroborating with its popular medicinal use. Previous studies conducted by the team of the present proposal focused on the genetic diversity and chemical composition of volatile (essential oils) and non-volatile (extracts) metabolites with 12 populations of L. pinaster occurring in the northern (2), southeast (1) and south (10) regions, showed significant a genetic diversity (microsatellite marker) between populations and in chemical composition. The essential oils and extracts of L. pinaster were also evaluated for their antiproliferative activity (3 populations: southeast and south) and antimicrobial activity (a southern population) against Streptococcus mutans, one of the main etiological agents of dental caries, with a positive result resulting in a patent of invention. The results obtained demonstrate the antitumor potential of L. pinaster essential oil. The present project aims at the continuity of studies on populations of L. pinaster extending the sampling regions, especially in the north and southeast regions, in order to attain greater representativeness of the chemical and genetic diversity of the species in the State of Minas Gerais. The characterization of genetic diversity will be performed by SNP molecular marker and GBS sequencing. The essential oils will be extracted by hydrodistillation and the extracts will be prepared cold with ethanol. The analysis of the chemical composition of essential oils will be carried out by one-dimensional gas chromatography coupled to mass spectrometry (GC-MS) and comprehensive two-dimensional chromatography coupled to mass spectrometry (GCxGC-MS) and the extracts, by high-performance liquid chromatography-mass spectrometry (HPLC-MS).
