Abstract

Feline immunodeficiency virus (FIV) infection is the one of most important infectious diseases of domestic cats. For this reason, and because it causes an acquired immunodeficiency syndrome (AIDS) similar to Human immunodeficiency virus (HIV) in humans, FIV has been widely studied. Nevertheless, there are only a few descriptions of FIV complete genome and viral quasispecies analysis, and none using Next-Generation sequencing (NGS). Based on this, the present project aims to use NGS to obtain FIV genome sequences and to perform viral quasispecies analysis. The coding region plus the 3' untranslated region (U3) of FIV genome will be amplified from proviral DNA of naturally infected cats. The amplicons will be submitted to NGS on MiSeq system (Illumina) and the sequences will be used to make de novo assembly of nearly complete FIV genome and viral quasispecies analysis. Until this moment, there is no description of FIV complete genome of domestic cats from Brazil, and even on GenBank only 7 sequences are available. Therefore, this project expected to standardize a method for amplification and sequencing of FIV genome in a simple way, allowing a broader analysis to increase the knowledge on the biology and evolution of the virus and virus-host interaction.
