Abstract

Due to the high prevalence of dental erosion found in recent years, therapies are sought to prevent the occurrence or inhibit the progress of this alteration. Proanthocyanidin is an agent currently studied because of its properties, which among others, involves remineralization, reduction of demineralization and biomodification of dentin, inhibiting matrix metalloproteinases (MMPs) that degrade the dentin collagen. Thus, the objective of this study is to evaluate the role of Proanthocyanidin-based agents (gel and toothpaste) in the prevention of tooth wear (enamel and dentin) and degradation of the organic matrix of the demineralized dentin. To do this, 6 subprojects will be carried out: Subproject 1 - will be evaluated the effect of fluoridated remineralizing agents applied on eroded dentin, treated previously or not with collagen degradation inhibitors (among them Proanthocyanidin), on the dentin erosion; Subproject 2- will be chemically evaluated the surface of enamel and dentin, sound and eroded, with and without treatment with preventive agents developed for caries and erosion (among them Proanthocyanidin); Subprojects 3 and 4 - will be evaluate in vitro and in situ, respectively, a Proanthocyanidin based dentifrice on the wear resistance of the dentin subjected to erosion associated with abrasion; Subproject 5- Proanthocyanidin gel will be evaluated in the minimization of dentin wear and possible inhibition of demineralized organic matrix degradation; Subproject 6- will evaluate the effect of Proanthocyanidin with and without the presence of acquired pellicle in the inhibition of enamel erosion. In subprojects 1, 3, 4 and 5 the dependent variable will be the wear evaluation by profilometry. In subproject 2, the specimens will be chemically evaluated using XPS and EDX. The specimens of subproject 5 will also be evaluated for the degree of degradation of the organic matrix demineralized by the ELISA method. In subproject 6, the response variable will be the percentage of loss of surface hardness. The data will be analyzed by simple or factorial ANOVA tests, followed by multiple comparison tests, in addition to Multiple Linear Regression models, adopting a significance level of 5%. (AU)
