Gene expression and protein synthesis analysis of dental pulp cells from deciduous teeth after the use of Low Intensity Laser.

Abstract

This study aimed to evaluate the effect of Low Intensity Laser on cell viability, cell proliferation, gene expression and protein synthesis of cells derived from the pulp of human deciduous teeth after the variation of the parameters of photobiomodulation. Cells samples will be obtained and grown in specific culture medium with Fetal Bovine Serum (FBS). After the fourth passage, the sample will be irradiated with low intensity laser according to the photobiomodulation protocol by varying the power and the irradiation time obtaining different energy densities (1.2 to 6.2 J / cm2). The control groups of each experiment will be treated under identical conditions, except that the laser device will be kept off during the experiment. Viability analysis (MTT assay) and cell proliferation (crystal violet assay) will be performed at 6, 12 and 24 hours after irradiation. The gene expression of angiogenic growth factors (VEGF-A, VEGF-D, VEGFR1, VEGFR2, FGF2, PLGF, PDGF, PECAM-1 and BMP-9) will be quantified in the cell lysate by RT-PCR in 6, 12, and 24 hours after irradiation. Protein synthesis of the same growth factors will be quantified in the supernatant and cell lysate by ELISA multiplex assay at 6, 12 and 24 hours post irradiation. The values in triplicate will be analyzed of each experiment. The means and standard deviation will be tested for normality test. If the data present normal distribution, intragroup and intergroup comparison will be performed by ANOVA at two criteria, followed by the Tukey test. If the data do not present normal distribution, the intragroup comparison will be performed by the Friedman Test and the intergroup comparison will be performed by the Kruskal-Wallis test. The significance level of 5% will be adopted for differences to be considered statistically significant. (AU)
