Influence of ceramic veneers cemented over teeth with and without finish line on the inflammatory mediators and microbiota of gingival crevicular fluid. Clinical, controlled, randomized, blind, prospective and longitudinal study using ELISA and RT-PCR

Abstract

Objective: quantify the presence of biomarkers and colonizing bacteria of the gingival sulcus, to verify periodontal clinical parameters and marginal adaptation of the natural teeth submitted to aesthetic treatment with laminate veneers with and without finish line determined by randomization and cervical margin placed until 0,5 mm in within the gingival sulcus. Materials and Methods: 28 patients will be treated with laminate veneers in monolithic lithium dissilicate with 0.2mm thick at the cervical area, which will be cemented onto prepared and unprepared maxillary anterior teeth. For all the groups, the crevicular gingival fluid (GCF) will be collected prior to restoration cementation (baseline) and at future times (t): t7 - 7 days, t15 - 15 days, t30 - 30 days, t60 - 60 days and t180 - 180 days after restorations cementation, where the GCF volume will be calculated and the follow biomarkers quantified by ELISA: TNF-alfa, IL-1beta, IL-6, MMP-8, MMP-9, collagen type I, TIMP-1, TIMP-2 and TGF-beta. At the same times, the presence of the bacteria S. sanguis, S. mitis, A. naeslundii, F. nucleatum, P. intermedia, P. gingivalis, T. forsythia e T. denticola will be quantified by use of real-time PCR. Clinically, will be evaluate the periodontal clinical parameters: plaque index (PI), bleeding on probing (BOP), probing depth (PD), clinical attachment level (CAL), gingival recession (GR), gingival width (GW) and gingival biotype (GB). Also, will be evaluate the marginal adaptation in all times of study. Replicas of cervical area will be made with epoxy resin at (t0) 7, (t2) 30, and (t4) 180 days after cementation to verify the marginal adaptation and overcontour in the scanning electron microscope (SEM). Results: After the tabulation of the data, the assumptions of normality and homogeneity will be verified, considering the application of ANOVA test with measures repeated in time. If the assumptions are not met, non-parametric tests will be performed. The significance level considered for difference between averages will be 5%. (AU)
