Abstract
Polycystic ovary syndrome (PCOS) is a complex disorder presenting reproductive and metabolic consequences. It has been associated with insulin resistance and hyperinsulinemia, which are responsible for the increase in metabolic risk and type II diabetes. However, it is estimated that 25-50% of PCOS patients do not present such abnormalities, demonstrating the heterogeneity of the syndrome. Studies with the objective to differentiate such patients from the ones with metabolic compromising are scare and have conflicting results. The aim of the present study is to differentiate patients with PCOS with and without glucose intolerance by analyzing clinical, laboratorial, epigenetics and metabolic characteristics. The primary research question would be: Is it possible to identify biological markers that inform about metabolic impairment in PCOS women? Clinical and anthropometrical data as well as blood samples will be collected from 60 patients (being 20 health controls, 20 PCOS with glucose intolerance and 20 PCOS without glucose intolerance). An expression panel of 84 selected plasma microRNAs related to the development of diabetes (Sabiosciences, Qiagen) will be analyzed by quantitative real time PCR (qRT-PCR). Furthermore, profile of 186 plasma metabolites (target metabolomics) will be analyzed by electrospray ionization (ESI) tandem mass spectrometry (MS/MS). All results will be submitted to statistics and bioinformatics analyzes. (AU)
