Evaluation of proteins differentially expressed in saliva associated with periodontitis in obese and eutrophic pregnant women before and after labor

Abstract

Obesity and pregnancy are factors in which systemic and oral changes can be noticed. The adipose tissue of obese patients secretes inflammatory mediators that reduce the immune response of the host and exacerbate the inflammatory response. The mechanism during pregnancy is similar, due to estrogen and progesterone levels alterations. Therefore, both obesity and pregnancy are associated with high inflammation in periodontal tissues. Salivary proteome analysis is a method for the large-scale identification of proteins and biomarkers, which can be used to diagnose local and systemic diseases. The aim of this study will be to to monitor the inflammatory status of obese women through the identification of proteins differentially expressed in saliva associated or not with periodontitis before and after labor. The sample will be consisted by 40 pregnant women divided into: GOP - Obese with periodontitis (n=10); GOWP - Obese without periodontitis (n=10); GEP - Eutrophic with periodontitis (n=10); and GEWP - Eutrophic without periodontitis (n=10), who will be evaluated in the 3rd trimester of pregnancy (T1) and after, at least, 3 months of delivery (T2). Grouping sample will occur according to anthropometric parameters - weight, height and Body Mass Index (BMI); and periodontal parameters - Periodontal Pocket Depth (PPD) and Clinical Attachment Level (CAL). Periodontitis will be considered from the presence of 2 or more interproximal sites with CAL e 3 mm and 2 or more interproximal sites of different teeth with PPD e 4 mm or 1 interproximal site with PPD e 5 mm. For proteomics salivary analysis, the sample will be evaluated individually and, after, they will be processed for analysis by Nano Liquid Chromatography Electron Spray Ionization Tandem Mass Spectrometry (nLC-ESI-MS/MS). The obtained MS/MS spectra will be compared to human protein databases (UniProtKB/Swiss-Prot). Free marker quantification will be performed using Protein Lynx Global Service (PLGS) software. For the comparison of the anthropometric and periodontal parameters, the Kolmogorov-Smirnov test will be applied to evaluate the normality of the quantitative variables (weight, height, BMI, PPD and CAL). ANOVA will be applied for the quantitative variables with normal distribution and Friedman for the quantitative variables without normality. The level of significance of 5% will be adopted. (AU)
