Abstract

Zucchinni yellow mosaic virus (ZYMV) of melon is controlled with resistant varieties. The access PI41427 is the principal source of resistance used in breeding programs and its resistance is controlled by the major dominant locus Zym-1, which is closely linked to the microsatellite marker CMAG36 located in the linkage group II. The literature indicates the possibility of the existence of at least another resistance locus in this access, called Zym-2, with a less pronounced phenotypic effect and a possible epistatic effect with alleles of Zym-1. Evidence from studies conducted in our lab with low-resolution genetic maps indicates a probable location of Zym-2 in linkage group X. The availability of the genome sequence of melon brought the perspective to identify the genomic locations of these two loci and the resistance genes by analyzing their genic context. Thus, the objective of this project is to align the sequences of chromosomes 2 and 10 with the linkage groups II and X and to analyze the composition and the expression of genes located in the Zym-1 and Zym-2 loci in an attempt to identify the resistance genes to ZYMV. However, in the case of Zym-2, linkage analysis with a greater number of microsatellite markers and a segregating population of 500 F2 plants from the cross `PI414273´ x `Védrantais` will be needed. In addition to these objectives, the project will also characterize the resistance of `PI414273´ with respect to the biology of the virus. For this, a methodology to quantify the virus by qPCR will be developed and used to determine the effects of the resistance alleles of both loci on the replication and translocation of ZYMV. (AU)
