The development of computational methods for analyzing modular transcriptional repertoires has proven to be a fundamental tool for uncovering the immune defense mechanisms associated with good and bad responses to respiratory viruses. Our group adopted this approach for studying the processes of development and demise in the immune system (thymus) and also for investigating the immune response to respiratory viruses. Recently we were able to demonstrate that in peripheral blood monocytes there are distinct transcriptional modules associated with the responses to rhinovirus or to syncytial respiratory virus in children under six months hospitalized with bronchiolitis. Now the same methodology will be used for investigating the responses of adult and pediatric patients to SARS-CoV-2. The study of the differences between adult and pediatric patients regarding the immune response and disease course constitute a unique opportunity for developing new therapies enormously demanded for avoiding the health system collapse now and in the post-pandemic period (2022-2024), as it was shown by recent epidemiological studies. Therefore, this project is centered in the comparative study of the leukocyte transcriptional response to the infection by SARS-CoV-2 in adults and children, and in oligosymptomatic and severe cases as well.
