Abstract
Mammary tumors represent the most common neoplasms in dogs and women both. Similarities between the tumors in both species include spontaneous appearance, tumor evolution, biological behavior, and the expression different molecules. Thus, the dog is considered a natural model for the disease. Some tumors represent a therapeutic challenge and new therapies are needed for better patient prognosis. Different chemotherapeutic drugs were extracted from botanical compounds, and the Brazilian savanna presents a diversity of plants that may have potential for identification of new drugs. Synadenium grantii Hook F., a botanical species used in ethnomedicine in central-western and southern Brazil, is used empirically to treat a variety of diseases, including cancer. Thus, studies with S. grantii latex are promising for identification of antitumor compounds. Thus, the present research aims to evaluate chemical characteristics of the crude ethanolic extract (BSE) of Synadenium grantii Hook F. latex, to separate and identify active compounds by high performance liquid chromatography (HPLC) and high resolution mass spectrophotometry (EMAR). Also, to evaluate the effects of crude ethanolic extract of S. grantii latex in vitro and in vivo models of breast carcinoma. The HPLC will be performed to identify the compounds present in S. grantii latex, and their in vitro antitumor action will be evaluated in female and female mammary tumor cells. For this, the IC50 dose of S. grantii latex will be identified, and the cells will be treated to evaluate the effect of latex on cell migration and induction of apoptosis/necrosis. Subsequently, the in vivo antitumor effect will be evaluated on ET. Therefore, the in vivo TE model will be established to evaluate the antitumor effect of S. grantii latex. (AU)
