Genetic mapping and high-resolution genotyping of resistance loci associated to Fusarium oxysporum f. sp. phaseoli in common bean

Abstract

Among the limiting factors that contribute to the low productivity of the common bean, the crop is diseases of varied etiology. Fusarium wilt stands out for being considered one of the main soil diseases in the crop, is caused by the fungus Fusarium oxysporum f. sp. phaseoli (Fop). Mainly due to the expansion of the culture in irrigated systems, with successive plantings of beans, the disease has spread and deserved greater prominence in recent years. To obtain bean varieties with resistance to such pathogens, it is necessary to know the genetic architecture of the resistance to this pathosystem in common beans. In this project, we aim to validate the loci found by GWAS using a Mesoamerican panel of the BAG/IAC in an F2 population from the crossing of contrasting genotypes for Fop resistance, with two isolates (Fop UFV01 and Fop 18001) and two evaluation parameters (SEV and AUDPC). SNPs generated by BARCBean12K technology will be used for genotyping, and after selecting putative candidate genes, we aim to carry out high-efficiency genotyping with the TaqMan and KASP (Kompetitive Allele-Specific PCR) systems. It is hoped that this study can point out some efficient markers to be used as a tool for marker-assisted selection in obtaining elite cultivars aiming at resistance to Fusarium wilt. (AU)
