Effect of experimental dentifrices with epigallocatechin 3-gallate and chitosan on dentin hypersensitivity: in vitro study.

Abstract

Dentin hypersensitivity is a frequent clinical condition that is characterized by pain or discomfort caused by thermal or osmotic stimuli. Daily oral hygiene is important to prevent or minimize dentinal hypersensitivity, and the use of a dentifrice that is able to occlude dentinal tubules and resist everyday acid challenges can be a useful strategy in the prevention or treatment of dentinal hypersensitivity. The aim of this study will be to investigate in vitro the effect of applying experimental dentifrices containing epigallocatechin 3-gallate (EGCG) and chitosan on dentin that simulates hypersensitive areas. For the study, 75 specimens of bovine root dentin will be used, which will initially be immersed in EDTA 0.5M to open the dentinal tubules. Later these will be divided into 5 groups: Base dentifrice; Toothpaste containing fluoride; Toothpaste containing EGCG; Toothpaste containing chitosan nanoparticles; Toothpaste containing chitosan nanoparticles encapsulated with EGCG. The specimens will undergo a cycle model of erosion and treatment for 5 days. The erosion will consist of immersion in citric acid (2 minutes, 0.3%, pH 2.6, 4 times a day), and the treatment will be carried out by brushing with slurry of each of the toothpastes (2 N, 45 strokes). Quantitative response variables will be obtained through analysis of root dentin permeability and confocal laser microscopy (evaluating the increase/decrease in the number, area and perimeter of dentinal tubules). The qualitative variable will be performed through the analysis of scanning electron microscopy. The results obtained will be analyzed. If there is homogeneity of variance and normal distribution of data, a parametric test will be performed using Analysis of Variance, otherwise, a non- parametric test will be used. Tests will also be performed to correlate the data analyzed by confocal microscopy with the final values of dentin permeability. (AU)
