Abstract
The main goal of this proposal is to obtain funding to acquire equipment for the creation of a Multiuser Facility for Metabolic, Cardiovascular and Respiratory Phenotyping of rodents in different ambient temperatures and light-dark cycles. The funding will be destined for the acquisition of the Promethium Core (Sable Systems) and climate ambient chambers (4 chambers) coupled to a system of telemetry for mice and rats. This modern system will allow the simultaneous evaluation of several rodent physiological parameters such as oxygen consumption, carbon dioxide production, locomotor activity, water intake, food intake, body weight, basal energy expenditure, core body temperature, brown adipose tissue temperature, thermogenic capacity, heart rate, and blood pressure. In addition, these assays may be performed in different ambient temperatures (varying from 4 to 40°C) and distinct combinations of light-dark cycle, meeting the most diverse experimentation needs. The physiological variables mentioned above could be evaluated in a large number of rats or mice (120 animals simultaneously), including immunosuppressed and pathogen-free mice. This Facility will allow the execution of highly complex experiments, benefitting projects by students and researchers, laboratories and companies that investigate the regulation of body weight, body composition, adiposity, energy balance, water intake, food intake, metabolism, inflammation, immune response, respiration, acid-basis balance, glucose homeostasis, body temperature, blood pressure, circadian rhythms, among others. Furthermore, we will be able to perform pre-clinical studies for pharmaceutical industries. Therefore, it is evident that this Facility will potentiate the generation of a large amount of high-quality knowledge, increasing our national and international competitiveness, and accelerating the development of new and more efficient strategies to prevent and treat chronic metabolic diseases. This Facility will also meet our present and future demands of ideal experimental conditions of temperature and photoperiod regimen. Noteworthy, this Facility may contribute to innovation activities through contracts for the provision of services and scientific cooperation, which may be established with industries. (AU)
