EMU: Acquisition of BD FACSymphony A5 equipment for multiparametric flow cytometry

Abstract

The BD FACSymphony A5 is a cell analyzer that uses the principle of flow cytometry. The equipment has 3 lasers: blue (488nm), red (640nm) and violet (405nm). All three lasers will have a power equal to 100mW and will allow the operator to control them individually via software (on, off and power adjustments). The instrument will be equipped with 21 independent detectors, allowing 21 different cellular characteristics to be evaluated from each of the thousands of cells capable of analysis per second. The photodetectors, 1 of the photodiode type and 20 of the photomultiplier tube type, were accommodated in such a way as to allow the evaluation of two directions of light dispersion produced by the blue laser (FSC and SSC), in addition to 19 fluorescences, 7 of which were generated by the blue laser. , 3 by the red laser and 9 by the violet laser. The distribution of detectors and the personalized choice of each of the optical filters were carried out in order to allow the best possible combination of specific fluorescence for each of the three lasers, without the need to apply mathematical methods of spectral deconvolution. The present proposal aims to acquire equipment of extreme importance for science, technology and innovation, aimed at immuno-oncology. Such equipment is essential for cutting-edge research involving immunological markers. However, Hospital de Amor de Barretos does not yet have such equipment and this acquisition would also benefit other institutions in the region, which will be able to take advantage of the institution's multi-user structure.
