Neurochemical control of body temperature

Abstract

Body temperature (Tb) is one of the vital signs continuously monitored in all hospitalized patients, along with respiratory and cardiovascular parameters. This emphasis in Tb is given because on its influence on physiological functions and also because alterations of Tb are important signs of diseases. Tb alterations are observed during fever and anapyrexia, and during septic shock where the organism seems to lose its ability to contrai Tb. Fever and anapyrexia are regulated increase and decrease of Tb, respectively, and septic shock is characterized by a systemic inflamation response to infection (sepsis) accompained by hypotension and secondary clinical manifestations which include fever or hypothermia. A number of studies have assessed the mechanisms involved in these responses and data acumulated suggest that the central nervous system (CNS) is an important site for a number of hormones and neuromodulators. Thus, the present proposal will focus on the study SNC. The aims of this proposal are divied in 8 sections: 1) verify the involvement of ionotropic glutamate receptors in hypoxia-induced hyperventilation and anapyrexia; 2) verify the involvement of nitric oxide and carbon monoxide in platelet derived growth factor- induced fever; 3) verify role of locus coeruleus in LPS-induced fever; 4) verify the role of serotonin, in the CNS, in hypoxia induced hyperventilation and anapyrexia; 5) verify if adenosine and/or dopamine are endogenous neurotransmitters in hypoxia induced anapyrexia; 6) verify if there is difference between anapyretic responses in dark and light phases and if melatonin participates in this response; 7) verify if AVP deficiency during experimental septic shock is due to NO inhibitory effect on AVP release and verify if AVP depletion in the pituitary is associated with experimental septic shock development; 8) verify the effects of AV3V electrolytic lesion and OVLT chemical lesion on AVP and ocitocin release in septic shock.
