Analysis of the influence of fluoride in bone repair in dental alveoli in rats

Abstract

The comprehension of the molecular mechanisms involved in bone repair could help in the treatment of bone defects. Fluoride is an element daily present for most people, used in drinking water, dentifrices and also in the therapy of osteoporosis. In such case it is important the understanding of the effect of fluoride in physiologic repair in bone tissue. This study will use four groups of five 80-day-old Wistar rats each wich will receive drinking water containing the following single doses of fluoride: 0 (control), 5, 15 and 50 ppm F, during 60 days. The incisors of the animals will be extracted. After these treatments, the animals will be euthanized at time zero and after 7, 14, 21 and 30 days post-surgical. Dental alveoli will be collected for microscopic (stained by hematoxylin eosin and immunohistochemical), fluoride concentration, Western blotting (BMPs, MMPs, TRAP, RANK, RANKL and OPG) and proteomic analyses . Fluoride in reaction tissues will be analyzed with the electrode, after hexamethyldisiloxane-facilitated diffusion. Proteins in reactions tissues will be profiled by two-dimensional gel electrophoresis. Gels from control and treated groups will be digitalized using the ImageScanner and analyzed with ImageMaster 2D Platinum software. Spots of interest will be excised for protein identification by mass spectrometry (MALTI-TOF). The effect of fluoride on the activity of protein phosphatases of bone cells and the phospho-tyrosin proteins will be also determined by Western blotting. Parametric analysis will be realized by analysis of variance (Tukey’s test if p<0.05). (AU)
