Studies regarding the sexual transmission of Toxoplasma gondii in goats (Capra hircus)

Abstract

In the reviewed literature, certain works reported the isolation of Toxoplasma gondii from semen samples of reproductive goats; however, up to the present date, none have determined its sexual transmission. To this aim, research will be realized using male goats of reproductive age inoculated with Toxoplasma gondii oocysts (via oral) or tachyzoites (subcutaneous via) from P and RH strains, respectively. Following the inoculation of the reproductive males, nonpregnant reproductive female goats with no infectious infirmities, including toxoplasmosis, will be synchronized for fertilization, so that the experimentally infected males can perform natural copulation. These will be divided into specific groups according to infection by oocysts or tachyzoites. Parasite presence in the goats will be studied by indirect immunofluorescent antibody test (IIFA) from the gestation period through to delivery. Next, the goats will be euthanized and tissue parasitism will be studied by bioassay, PCR and immunohistochemistry on fragments of skeletal and heart muscle, brain/cerebellum, retina, spleen, liver, uterus, vagina, ovary and placenta. Tissue parasitism will also be studied in the offspring using the same means. As a secondary objective, experimentally infected males will be studied regarding the presence of the protozoan in the serum, semen and tissues by IIFA. Thus, this research project aims to evaluate the possible sexual transmission of toxoplasmosis by coitus and the consequent dissemination of the infection in females and offspring. (AU)
