Abstract

Denture stomatitis, the most frequent lesion in elderly removable prothesis wearers, is characterized by an inflammation of the denture-bearing mucosa, especially underlying maxillary prostheses. It is related with the presence of Candida albicans (C. albicans) and local and systemic factors, such as the impaired immune response, which is common in elderly people. Monocytes play an important role in the development of a protective immune response against fungal infections since they release cytokines which recruit and activate leukocytes, collaborating to the fungal clearance. There are no literature data regarding the immunomodulatory function of monocytes by elderly people presenting denture stomatitis. Therefore, the aim of the present study is to evaluate the activation and production of pro- and anti-inflammatory cytokines by monocytes isolated from peripheral blood, challenged in vitro with C. albicans, in elderly people, wearing complete maxillary denture, with or not denture stomatitis, in comparison to elderly people non-denture wearers. Peripheral blood will be obtained from volunteer donors and the monocytes will be purified and cultured in culture plates presenting glass coverslip inside the wells, with and without lipopolysaccharide (LPS) or viable C. albicans ATCC 10231. After incubation for 18 hours, supernatants will be harvested for measurement of the pro-inflammatory cytokines interleukin-1 alpha (IL-1±), interleukin-1 beta (IL-1²), IL-6, IL-8, monocyte chemoattractant protein (MCP-1) and tumor necrosis factor-alpha (TNF-±), and the anti-inflammatory cytokines IL-10 and transforming growth factor-beta (TGF-²), by enzyme immunoassay (ELISA).The expression of the monocyte marker, CD-14, and MCP-1 receptor and granulocyte-monocyte colony-stimulating factor (GM-CSF) receptor will be evaluated in the coverslip presenting adherent monocytes, by means of immunofluorescence. The results will be expressed as mean ± standard deviation (SD) of the values obtained by each group and the statistical analysis will be performed using analysis of variance (ANOVA), followed by the Tukey test. P values less than 0.05 will be considered statistically significant. (AU)
