Use of bacteriological isolation, immunohistochemistry, PCR and histopathology by hematoxylin/eosin and Warthin-Starry stainning for the diagnosis of leptospirosis in aborted bovine fetuses

Abstract

Leptospirosis is a disease that causes abortion and low reproductive rates in cattle. Serological surveys in Brazil revealed high rates of seropositivity in herds with a wide territorial distribution, as well as isolation in aborted fetuses has been reported. This bacterium is fastidious and its isolation doesn't apply in frozen or autolyzed material. The Warthin-Starry (WS) staining reveals the presence of Leptospira spp dinned by silver, it is a technique easy and quickly to do in histological slides, but histopathology is affected by autolysis and freezing. Immunohistochemistry (IHC) and PCR are diagnosis that present high specificity and sensitivity, and have the advantage to be performed in frozen or in initial stage of autolysis. The aim of this project is to evaluate the occurrence of Leptospira spp by IHC, PCR and WS staining, associated to the bacterial isolation in liver and kidney of aborted bovine fetuses sent to the "Centro de Pesquisa e Desenvolvimento de Sanidade Animal do Instituto Biológico" for differential diagnosis of abortion. The microscopic lesions caused by the agent will be studied in histopathological slides of liver, kidneys, heart, lungs, spleen and brain stained by hematoxilin/eosin (HE), because there are few histopathological descriptions in aborted bovine fetuses published.
