Abstract
The fungi Paracoccidioides brasiliensis and Sporothrix schenckii are of a great interest in medical and biological areas. The first is the agent of paracoccidioidomycosis, an endemic disease in many regions of Latin America, mainly in Brazil. The second is the etiological agent of sporotrichosis, subcutaneous mycosis, a re-emergent disease in our environment, and nowadays constitutes a zoonosis, because once infects cats and they are transmitters to humans. The proteomics, cromossomics and biochemicals characteristics of these fungi are not so studied. Although, in our study we will focus these aspects in these two agents for a better understanding of its biology. In the S. schenckii complex we will look for molecular evidences of intra-specific variations, through proteomic differential analysis among the species S. brasiliensis, S. globosa, S. Mexicana and S. schenckii, availing the differentially expressed protein and also, through immunoproteomic to determine the immunogenic proteins in the fix and lymphocutaneous sporotrichosis types. Also, in relation to S. schenckii, the variation of chromosomal profiles from different strains will be studied, mapping some of gene. Proteomic profiles of different P. brasiliensis isolates will be studied, and through immunoproteomic, the reactive proteomic map related with recognizing specifics antibodies from patients on the stage of acute and chronic paracoccidioidomycosis will be determined, as well as the characterization of a calnexin by the detailed study of its cellular location during the dimorphic transition and after stress conditions.
