Abstract

Capillary electrophoresis is an analytical technique of separation based on differences between the mobilities of charged species in an electrolyte solution that can be aqueous or organic, inside a small capillary under the influence of an electric field. Among the main advantages of capillary electrophoresis can be cited: the high efficiency, versatility, the demand for small sample volumes, reduced analysis time, the minimum consumption of solvents, the low cost of material consumption, the ability to interface with various detection systems, and simplicity of the basic instrumentation. Thus, due to efficiency, selectivity and versatility, this project proposes to demonstrate the application of this technique in different areas. So, is being proposed the enantioselective analysis of fluvastatin and its major metabolite in plasma, since there is still no method described in the literature for the analysis of this drug by capillary electrophoresis. In other applications, two macromolecules will be considered: the interferon-± (INF-±) and ±-1acid glycoprotein (±1-AGP). The analysis of INF-±, a biopharmaceutical, will be held in different commercial drugs with application in quality control of biopharmaceuticals and the analysis of ± 1-AGP, a biomarker will be held in the plasma of patients admitted to HCFMRP/USP with the objective of relate the concentration and structure of this glycoprotein in different developmental stages of the inflammatory state. Finally, it is proposed the development and validation of a method for analysis of imatinib mesylate and its metabolites in plasma. The method will be applied on plasma samples from patients with chronic myeloid leukemia treatment, to quantify the drug and help in understanding the mechanism of resistance observed in some patients.
