The influence of Diabetes Mellitus in repair of autogenous bone graft covered or not by reabsorbable collagen membrane

Abstract

This aim of the present study is to describe qualitatively and quantitatively the bone healing process of autogenous bone graft onlay, with or without resorbable collagen membrane; analyze by immunohistochemistry, the influence of angiogenesis in the process of bone resorption of autogenous bone graft onlay and to correlate these findings with the presence of diabetes mellitus to assess the effect of disease on guided bone regeneration. For this, it will be used 60 adult rats (Rattus norvegicus, Albinus variation, Wistar) ninety days old, weighing around 300 grams, which will be divided randomly into two experimental groups, each one containing 30 animals: Group Test compound rats with diabetes induced by alloxan administration; control group composed of normoglycemic rats. It will be considered diabetic animals those showing blood glucose above 200 mg / dl. All animals will be subjected to grafts in the right jaw and left. The grafts in the left lower jaw will be covered with resorbable collagen membrane, while the grafts in the mandible on the right side will be held in the same way, but without the covering membrane collagen. The animals will be euthanized by cardiac perfusion during periods 0 hours, 7, 14, 21, 45 and 60 days. Histomorphometric analysis will be performed and the data will be subjected to analysis of variance (ANOVA) and Tukey test, both at the conventional significance level of 5%. The immunohistochemical markers used are RANK, RANKL and Osteoprotegerin, for assessment of bone resorption. For analysis of angiogenesis, the markers will be used Tranglutaminase II and VE. (AU)
