Study of imunnomodulatory and antimicrobial anti-Candida actions of Lactobacillus casei

Abstract

Probiotics are dietary supplements constituted of live microorganisms or microbial cells components that are benefic to the health of the host. Since probiotics consumption seems to assist in eliminating of yeasts of Candida genus and in recovering of immune balance, this work aims to study, in vivo and in vitro, the immunomodulatory and antimicrobial anti-Candida actions of Lactobacillus casei, considered a probiotic species. For this, suspensions of Candida will be inoculated in the oral cavity of experimentally immunosuppressed mice for induction of candidosis. The animals shall be divided into four groups: the) with induction of candidosis without consuming probiotic, b) with induction of candidosis, with consumption of probiotic since 14 days before inoculation of Candida, c) with induction of candidosis with probiotic consumption during the inoculation of Candida, d) without inducing candidosis with probiotic consumption throughout the period of experimentation. Development and recovery of disease shall be histologically investigated through collecting and counting of yeasts in the oral mucosa. The presence of cells TCD4 +, TCD8 + and NKT will be investigated by Immunohistochemistry. In addition, serum samples will be collected from animals to analyze of nitric oxide, using Griess reagent, and INF-gamma, TNF-±, ² IL-1, IL-4, IL-6, IL-10, IL-12 and total immunoglobulins, by ELISA technique. Antimicrobial action anti-Candida of L. casei also will be investigated in vitro, using: agar diffusion, planctonic growth inhibition, inhibition of adhesion on epithelial cells and inhibition of the formation of biofilm. The ability of L. casei to induce the release of TNF-±, ² IL-1, IL-4, IL-6, IL-10, IL-12 and nitric oxide by macrophages (RAW 264.7) will be also investigated. (AU)
