Abstract

Colorectal cancer ranks third worldwide in incidence in both sexes and the second leading cause in developed countries. The preventive treatment directed to the consumption of foods that have anti-neoplastic, may help in reducing these rates. The grape is a fruit widely investigated in the prevention of cardiovascular disease and recently in research on cancer. The amount of phenolics present in grape has multiple biological effects such as antioxidant activity, anti-inflammatory antimicrobial and anticarcinogenic activities. Resveratrol one of the substances existing in this fruit has anti-inflammatory properties and may act in the inhibition of NF-kappa B, TNF-alfa and iNOS. This situation leads us to an investigation to detect the effectiveness of grape juice concentrate in colorectal cancer, dysplasia and cancer already present. The intent of this experimental research is to show the effectiveness of possible properties exist in colorectal cancer in the grape. Objective: To evaluate the effects of grape syrup in colon cancer induced in Wistar rats, by studying the expression of NF-kappa B, TNF-alfa and iNOS. Methods: Will be used 60 male rats aged 5-7 weeks weighing approximately 200-280g were divided into six groups (n = 10) animals each, where the SHAM control group will receive saline. In the experimental group will receive all of azoxymethane (AOM) 15mg/kg body weight for one day a week for 2 weeks and divided into four groups: G-O2 that after receiving AOM not receive treatment, G-03 will receive a standard diet over the grape syrup 2% every two days two weeks before receiving the first injection of AOM and continue collecting until the end of the experiment, G-04 1 week after the last application of AOM is treated with 1% grape syrup every 2 days; G-05 1 week after the last injection of AOM is treated with 1% grape syrup every 2 days, G-06 will be treated with grape syrup to 2% of AOM without receiving application. The expression of NF-kappa B, TNF-± and iNOS will be studied by reverse transcriptase (RT-PCR) and real-time PCR (qPCR). The endogenous genes GAPDH and beta-actin will be used for control. (AU)
