In vitro expression of adhesion molecules on in situ like malignant areas

Abstract

In order to occur the neoplastic progression, it is required the occurrence of several genetic and epigenetic events, including the overexpression of molecules that regulate cell growth, proliferation, motility and invasion to surround tissues, as well as cell surface receptors and adhesion molecules alterations which regulate cell-cell and cell-extracellular matrix interactions. Some studies have emphasized the importance of myoepithelial cells in the neoplasms behavior, highlighting its role as a tumor suppressor by promoting important defense against the cancer invasion. Considering an in vitro model of the tumorigenesis process and based in our previous studies, the aim of this study is to analyse cell-cell and cell-matrix interactions on sites that mimic an in situ situation when malignant squamous cell are surrounded by benign myoepithelial cells from pleomorphic adenoma, under the influence of fibronectin substratum at different periods of cell culture. The purpose of this investigation is identifying the moment of integrins (alpha5beta1, alpha5beta6), E-cadherin and/beta-catenin expression during the neoplastic process by indirect immunofluorescence.
