Effects of low level laser on regulatory myogenic factors expression and matrix metalloprotease 2 activity during skeletal muscle repair following cryolesion in rats

Abstract

Low level laser therapy (LLLT) is widely used in order to promote faster and better quality repair muscle after different types of injury, however, many studies show controversies in the literature with regard to parameters. The aim of the present study is to determine the effect of LLLT on the expression of myogenic regulatory factors (MyoD, Myf5, myogenin and MRF4) and on matrix metalloprotease 2 (MMP2) activity in the tibialis anterior (TA) muscle of rats following cryoinjury. Methods: Adult male Wistar rats (n=65) will be randomly divided into five groups: (1) control (no lesion, untreated,; (2) Sham group (without lesion and muscle will be only exposed); (3) cryoinjury group euthanized after 1, 3 and 7 days; (4) cryoinjury LLLT group euthanized after 1, 3 and 7 days; (5) without lesion euthanized after 1, 3 and 7 days. Laser irradiation will be performed three times a week on the injured region using the AsGaAl laser (780 nm; beam spot of 0.04 cm2, output power of 40 mW, energy density of 10 J/cm2, 10-second exposure time). The muscles will be removed and to isolate the total RNA using TRIzol reagent and to obtain cDNA synthesis. Real-time polymerase chain reactions will be performed using primers for MyoD, Myf5, myogenin and MRF4 and GAPDH was used to normalize the data. In addition, MMP2 activity will analyze by zimography and its presence will be confirmed by Western Blotting. (AU)
