Influence of diet supplementation of rosemary (Rosmarinus oficinalis L.) on cardiac remodeling in rats subjected to acute myocardial infarction

Abstract

Cardiac remodeling has a key role in ventricular dysfunction after myocardial infarction. Some cellular mechanisms such as oxidative stress, inflammation and fibrosis modulate this process. Thus, treatment or repair of post-infarction changes has great clinical relevance. There is great interest in natural products with antioxidant properties, especially rosemary. Objective: To analyze the influence of the consumption of dietary rosemary extract on cardiac remodeling after acute myocardial infarction (AMI) in rats. Methods: Wistar male rats with 200-250g will be allocated in six groups: 1) Sham group fed cereal-based chow and not subjected to AMI (SA0, n = 30), 2) Sham group fed with chow supplemented with 0.02% rosemary extract (diet 1) and not subjected to AMI (SA1, n = 30), 3) Sham group fed cereal-based chow supplemented with 0.2% extract rosemary (diet 2) and not subjected to AMI (SA2, n = 20), 4) AMI + diet based on cereals (IA0, n = 30), 5) AMI + diet 1 (IA1, n = 30; 6 ) AMI + diet 2 (IA2, n = 30). After three months, echocardiography study and euthanasia of animals for collection of biological material will be done. Cardiac tissue will be evaluated: 1) morphometric study, 2) analysis of oxidative stress by SOD, catalase and GPx and peroxidation, superoxide production and NADPH oxidase activity, 3) analysis of Nrf-2, HO-1 and GPx, 4) apoptosis study, 5) evaluation of inflammatory cytokines and 6) activity of metalloproteinases. Comparisons will be made by two-way ANOVA. (AU)
