Abstract

Micro-organisms from extreme environments have been focus of great interest because its biodiversity and resistance to climatic and geographic conditions. Fungi from Antarctic environment requires special attention because they represent great potential to exploration active substances with specific properties for applications in R & D. The enzymes produced by these micro-organisms may have biological advantage in processes that require medium and low temperatures and for that, growing conditions should be adequate. In this context, this project aims to establish laboratory cultivation conditions, in order to select potential producing yeast proteases, and to evaluate different parameters of cultivation for the favoring protease activity. It will be used 90 previously isolated yeast (PDA medium at 15 ° C) in the Division of Microbial Resources - DRM CPQBA / UNICAMP, from different samples from Antarctic continent. The selection of yeast producing proteases will be made from plating on Sabouraud Agar with 10% of skim powder milk, at 25°C. From the protease positive yeasts, it will be performed cultivations in liquid medium (Sabouraud broth), in order to select the best protease activity. The yeast selected will be used to stablish the best growing conditions for favoring protease activity, using experimental design with the factors temperature, pH, NaCI concentration and nitrogen source (peptone, rice bran and ammonium sulphate) in medium containing glucose as carbon source. The experimental conditions stablished in synthetic medium, except glucose, will be used to evaluate protease's production by selected yeast, using sugarcane bagasse hemicellulosic hydrolysate. (AU)
