Developmente of specific immune response in calves and its influence on response to vaccine bovine viral diarrhea virus

Abstract

The aim of this study is to monitor the development of specific immune system of young cattle, as well as check their interrelation with passive and active immunization, using as model the bovine viral diarrhea virus (BVDV). For this, we used 20 calves distributed in four experimental groups (G1 to G3), composed of five calves. The animals in groups G1 to G4 are immunized with commercial vaccine containing strains of BVDV NADL and SINGER type 01 and strain 125 of BVDV type inactivated 02. The animals of the G4 will not be vaccinated (control). Calves will be primovacinados to 14 days after birth (pn) (G01) at 90 days pn (G02) and 180 days p.n. (G03). All calves will be revaccinated at 30 days after the first dose of vaccine. Blood samples of animals will be obtained in three different moments: before the 1st dose (T0), before the 2nd dose (T1) and 30 days after the last vaccine. Evaluations of the calves from G4 will be held at all times of harvest of animals belonging to the G1 to G3. To evaluate the immune response will be held the following evidence immunoassays, measurement of specific antibodies and cytokine blood by ELISA (Enzyme Linked Immuno Absorbent Assay) phenotype and activation of subpopulations of blood lymphocytes, CD4 +, CD8 +, gamma delta TCR +, CD21 + and CD25 + cells by flow cytometry, and IFN-³ production by blood mononuclear cells. Data distribution will be initially assessed by Kolgomorov-Smirnov test. If these data are parametric distribution will be initially submitted to analysis of variance (F test), which is significant it will confront the means by Duncan's test. Variables with non-parametric distribution will be compared using the Mann-Whitney test. The relationships between the variables will be studied by the correlation coefficient and linear regression, which will be submitted to variance analysis for verification of significance (AU)
