Abstract

The widespread use of topical and systemic antifungal agents as conventional treatment for oral candidosis has resulted in the development of resistance in C. albicans. Thus, it is necessary to develop alternative therapies for the treatment of oral candidosis. Histatin 5 is a human basic salivary peptide with strong fungicidal properties in vitro. This peptide is extremely rich in one or two specific amino acids, which are positively charged at physiological pH and inhibit biofilm formation. Thus, the aim of this study is to i) synthesize, purify and characterize Histatins-5, ii) determine of the minimum inhibitory concentration (MIC) of the agent on planktonic cells and biofilms of Candida albicans by XTT assay and by counting viable colonies, iii) determine the concentration and establish the appropriate immersion times required to promote the inactivation of micro-organisms using similar tests described above; iv) evaluate in vitro the cytotoxicity of this peptide using tests mitochondrial activity (MTT) and electron microscopy (SEM). Theses analysis will allow us the development of a protocol for prevention and treatment of candidiasis in denture users. (AU)
