Evaluation of antioxidant effects of 5-aminosalycylic acid, N-acetylcysteine, sucralfate, curcumin and aqueous extract of Ilex paraguariensis in contente and pattern of tissue expression of intercellular junctions proteins: study in rats

Abstract

chronic inflammation in the mucosa of the colon segments devoid of fecal stream. The disease presents clinical, endoscopic and histological features similar to inflammatory bowel disease suggesting common molecular basis. The emergence of the DC is related to a deficiency of short chain fatty acids (SCFAs) in the intestinal lumen, which represent the main energy substrate of the epithelial cells of the colon mucosa. A deficiency of intraluminal SCFAs alter the energy metabolism of cells of the colon mucosa, leading to the formation of oxygen free radicals (ROS). The colonic mucosa is particularly deficient in antioxidant defense systems. The imbalance between increased production of ROS and antioxidant deficiency provoke oxidative stress that cause tissue damage in the defense system that comprise the epithelial barrier of the colonic mucosa. The different systems of intercellular adhesion, mainly represented by systems of tight junctions and adherents junction are in one of the main mechanisms for maintaining the integrity of the epithelium colic. The rupture of the proteins that make up the intercellular junctions systems enables the invasion of the submucosal sterile layer by existing bacteria and antigens in the intestinal lumen, triggering the tissue inflammatory response that characterizes the disease. Although DC is a relatively common disease with increasing incidence worldwide, where oxidative stress due to changes in cellular energy metabolism seems the probable molecular mechanism of epithelial damage, to date not been studied experimentally the effects of substances with antioxidant activity, such as 5-aminosalicylic acid (mesalazine), n-acetylcysteine, sucralfate, curcumin and aqueous extract of Ilex paraguariensis, in tissue content and pattern of expression of protein constituents of of the intercellular adhesion systems in an experimental models of DC. The objective of this study is to evaluate experimentally the effects of mesalazine, N-acetylcysteine, sucralfate, curcumin and aqueous extract of Ilex paraguariensis in the maintenance of content and expression pattern of proteins in systems occlusion junctions and intercellular adhesion in an experimental model of colitis exclusion (AU)
