The study aims to evaluate the analgesia and physiological effects of continuous intravenous infusion (CRI) of morphine, burtophanol, methadone or tramadol in horses with lipopolysaccharide (LPS)-induced carpal synovitis. Twelve adult healthy horses (350 - 500kg) undergoing a synovitis model, induced by injecting 0.5 ng of LPS in the left or right radiocarpal joint, will be submitted to 2 of 4 intravenous treatments: Bolus of 0.15mg/kg followed by 0.1mg/kg/h of morphine (Mo); bolus of 25¼g/kg followed by 25¼g/kg/h of butorphanol (B); bolus of 0.5mg/kg followed by 0.5mg/kg/h of methadone (Me), and bolus of 1.5mg/kg followed by 1mg/kg/h of tramadol (T). The treatment started 3h after synovitis induction and CRI will be maintained for 3 h later. Lameness, heart rate, systolic arterial pressure, respiratory rate, temperature, and intestinal motility will be evaluated before LPS injection (baseline), and 1.5, 3, 4, 5, 6, 7, 8, 9, 10, 12, 14, 16 and 24h post-LPS. It is expected that one or more treatments may be effective in pain control of those horses and, due to the doses and the method of administration, it is possible that the adverse effects can be minimized, achieving considerable analgesia and physiological stability.
