Mesenchymal stem cells and platelet rich plasma associated with microfractures in the treatment of articular chondral defects in horses

Abstract

Osteoarthritis is one of the most prevalent causes of lameness in horses, and it is associated with poor performance, physical incapacity and early retirement of sport horses. Articular cartilage is the key target of the degenerative disorders of osteoarthritis. Nowadays studies have attempted to overcome barriers of the articular cartilage repair. The cell therapy and biomaterials used as supports in the standard therapies have contributed significantly not just for reduce the pain and joint deterioration, but even as an alternative treatment for regeneration. The mesenchymal stem cells (MSCs) have received a high importance in the treatment of osteoarthritis due to its therapeutic potential and a large immunomodulatory capacity. Similarly, the use of Platelet Rich Plasma (PRP) in animals with chondral defects has resulted in a significant decrease in pain and improvement in the rate of chondrocyte proliferation and extracellular matrix production. The use of microfractures, as a therapy already renowned and which has presented favorable results, applied in combination with the MSCs and PRP, which are already known to be beneficial, should still be investigated to enhance the articular cartilage repair. Therefore, the aim of this study is to compare the effect of treatment only with microfractures to the treatment with microfractures associated with the MSCs and PRP in articular cartilage defects experimentally induced by arthroscopy in the medial femoral trochlea of the patellofemoral joint of horses. This study attempts to an optimal approach to reducing the formation of fibrocartilage and reach the full recovery of the joint affected by osteoarthritis. (AU)
