Screening of plants from Brazilian Pantanal with antimicrobial activity against enterobacteria, Pseudomonas spp. and Staphylococcus spp.

Abstract

Pantanal biome has a great variety of vegetal species with potential medicinal use. Enterobacteria, Pseudomonas spp. and Staphylococcus spp. can be involved in periodontal diseases and/or endodontic infections. The aim of this study is to evaluate the antimicrobial effect of plants from Brazilian Pantanal against clinical enterobacteria, Pseudomonas spp. and Staphylococcus spp., to test the best extraction method of bioactive compounds of these plants and to obtain a chemical profile of the most active extracts. The extracts will be prepared from leaves collected during the flood season. The leaves will be dried and pulverized prior to the extract preparation. Sixty extracts (proportion powder/solvent 1:20 weight/volume) will be prepared varying the solvents, the temperature and/or time of extraction (Water: 100oC/5 min, 55oC/1h ou 25oC/72h; Ethanol: 70%/72 h or 99%/72 h; Hexane: 72 h). Initial screening will be done with reference strains Escherichia coli ATCC 23922, Pseudomonas aeruginosa ATCC 9027, Staphylococcus aureus ATCC 6538 e S. epidermidis ATCC 12228 through the agar well difusion test, using the extracts at 50 mg/ml. The most active extracts will be tested regarding minimum inhibitory concentration and minimum bactericidal concentration with the microdilution method in 96-well plates using the reference strains and 108 oral isolates from healthy volunteers (50 isolates from enterobacteria / Pseudomonas and 58 isoaltes from Staphylococcus spp). Data obtained will be statistically analysed using the software GraphPad Prism version 3.02. The extracts with the best antimicrobial activity will have their chemical profile determined using the hyphenated techniques. (AU)
