Influence of diferente irrigating solutions associated to photodinamic therapy: in vitro and in vivo study

Abstract

Recently, it has been showed strong evidence of the photodynamic therapy (PDT) success in microbial reduction. PDT is based on appropriate physical, chemical and biological effects, that occur after administration of a photosensitizing agent (PS) activated by visible light of specific wavelength, with intent to destroy the target cell. The aim of this study is to evaluate the behavior of PDT in comparison with some irrigating solutions, as well as the association between the irrigating solutions and PDT using different methodologies: 1-antimicrobial effects in vitro by direct contact test against strains of Enterococcus faecalis and Porphyromonas gingivalis. The results will be evaluated with ANOVA, Tukey and Bonferroni's tests (p <0.05), 2-cytotoxicity in L-929 fibroblasts by the MTT colorimetric method, 3-Production of cytokines (IL-1² and IL-6) by fibroblasts using enzyme immunoassay (ELISA). The results will be statistically analyzed by ANOVA with Bonferroni's correction (p <0.05), 4- tissue response to polyethylene tubes implant in the subcutaneous tissue of rats. The results will be statistically analyzed by Kruskal-Wallis test (p <0.05), 5-Production of cytokines (IL-1² and IL-6) by the tissue, analyzed by ELISA. The results will be statistically analyzed by ANOVA with Bonferroni's correction (p <0.05). (AU)
