The aim of this research is to evaluate the association among Casein phosphopeptide/Amorphous Calcium phosphate and/or lysozyme, lactoferrin and lactoperoxidase in the glass ionomer cement for dentin carious treatment. One hundred third permanent molars will be obtained and flat dentin surfaces will be realized. The samples will be sealed except the coronary dentin and carious lesions will be artificially created with a padronized strain of S. mutans ATCC25175. The samples will be incubated in anaerobioses by a bacteriological incubator at 37°C for 1 month. During this period of time, BHI will be replaced each 2 days. The dentin carious lesions will receive 5 different ways of treatment: group 1 (positive control): without treatment; group 2: glass ionomer cement (GIC); group 3: GIC + lysozyme 1%, lactoferrin 1% and lactoperoxidase 1% (LLL); group 4: GIC + Casein phosphopeptide/Amorphous Calcium phosphate 3% (CP/ACP) and group 5: GIC + CP/ACP +LLL. After experimental periods (24 hs, 1 month and 6 months) the sealing will be removed for microbial counting. The S. mutans counting will be performed before sealing of the carious tissue, after 24 hs, 1 month and 6 months. The results will be transformed in log10 and will be submitted to a descriptive analyses. Wilcoxon test will be used to compare the total number viable S. mutans before and after the experimental periods in the same group. The percentage of microbial reduction will be calculated and the comparison among the different groups will be realized by Kruskal-Wallis test. (AU)
