Evaluation of bone repair in the bone / implant interface in osteoporotic rats treated with raloxifene or alendronate: a histometrical, immunohistochemical, epifluorescence and biomecanical analysis

Abstract

The present project has the aim ot evaluate the bone repair process in the bone/implant interface of rats with induced osteoporosis and treated with raloxifene or alendronate through histometrical, immunohistochemical, epifluorescence and biomechanical analysis. To perform this study, 80 rats (Rattus novergicus albinus, Wistar), with the 200g weight were divided in 3 groups, according to the analysis that will be realized, and divided again in other 4 subgroups according to the induced osteoporosis and medical treatment (SHAM DN = sham animals and with normal diet. OVX ST = rats submited to bilateral ovariectomy and feeded with poor Calcium diet (osteoporotic rats) with no medical treatment; OVX RAL = osteoporotic rats treated with raloxifene; OVX ALE = osteoporotic rats treated with sodium alendronate). Each animal will receive 2 implants, each implant installed in each tibial metaphises. On right tibia, it will be installed an usinade surface implant and in the left tibia, it will be installed a treated surface modified. The eutanasia of the animals will be performed in the periods of 14, 42 and 60 days after the instalation of implants depending on the analysis to be performed, through anesthetics overdose. For the histometrical and immunohistochemical evaluation, 20 rats will be sacrified at 14 and them more 20 rats will be sacrified at 42 days after the implants instalation. For the epifluorescence analysis, 20 rats will be sacrified at 60 days after the instalation of the implants. For the biomechanical analysis (referse torque), 20 rats will be sacrified at 42 days after the implants instalation. All of the data will be submited to normality curve in order to determinate the most appropriate test (parametrical or non parametrical) to e applied. The significance level of 0,05 will be aplicated to all of the tests. (AU)
