Abstract

Micro RNA (miRNA) are important regulators of gene expression and its changes are related to the development and progression of cancer. Studies of literature and previous results obtained by our laboratory imply some miRNA in progression and biochemical recurrence of prostate cancer. Are already available miRNA molecules to treat diseases. The purpose of this study is the research of the paper of miRNA in the treatment of metastatic prostate cancer models in vitro and in vivo. Method: The lines PC3, DU145 and LNCaP will be used in in vitro experiments. The miRNA and its antagomirs will be transfected using the lipofectamine and after 24 h of exposure will be evaluated proliferation rates, apoptosis, and aneuploidy by flow cytometry. Changes in cellular invasiveness will be analyzed using matrigel. After the experiments will be analyzed the expression levels of target genes RAS, MYC, MMP9 and CD44 by the technique of quantitative PCR in real time.Athymic mice, male with 5 or 6 weeks will be injected intraventricularly with line PC-3M-Luc-C6. On day 20 will be assessed development of bone metastases using analytical instrument IVIS imaging in vivo. The animals will be treated with the miRNA and its antiMiR in days 23 and 26. They will be evaluated by measuring the responses of the lesions with the same equipment on days 30, 34, 38, 42 and 44. On day 48 after the last imaging test the animals will be sacrificed and their organs analyzed macroscopically and microscopically. (AU)
