Antimicrobial and immunological role of cationic peptides in the prevention of early childhood caries and a proposal for incorporation of these peptides in nanoparticles systems as anticaries agents

Abstract

Early childhood caries (ECC) is a microbial infection that can severely compromises the dentition of young children. The etiology of ECC includes infection with selected bacteria, and dietary and host factors. In the oral cavity, the components of the innate immune system like antimicrobial cationic peptides (AMCP) are responsible for the maintenance of the ecological balance and the lack of opportunistic infections. Studies have demonstrated that children with ECC have low levels of some AMCP when compared to caries free children. The incorporation of fragments of their peptides in delivery devices could be an interesting way to prevent severe forms of caries in immunologically susceptible children. The objectives of this study will be to evaluate: 1) the in vivo influence of levels of putative cariogenic microorganisms and immunological factors on development of ECC; 2) the in vitro antimicrobial action of synthetic AMCP isolated/combined incorporated or not in nanoparticles systems. In part 1, biofilm will be collected from caries-active and caries-free children for putative cariogenic microorganism counts by Real Time-PCR. The quantification of AMCP using ELISA also will be quantified from saliva samples. In part 2, ATCC and clinical strains will be used to evaluate microbiological functions of synthetic AMCP, MIC/MBC and biofilm assays tests will be developed. The same microbiological tests will be performed for nanoparticles systems containing isolated or combined AMCP, besides toxicity tests on epithelial cells. Data will be statistically analyzed considering p d 0.05. This study intends to contribute to evaluation of the antimicrobial and immunological role of AMCP in the caries prevention and introduce a proposal for incorporation of these peptides in nanoparticle systems as anticaries agents.
