Mannanoligosaccharides in preventive medicine: studies in vitro and in vivo

Abstract

The aim of the research project will evaluate three different mannanoligosaccharides (MOS), Bio-MosTM, ActigenTM and ImmunoWallTM in Preventive Medicine, through studies in vitro: the ability of MOS agglutination in isolated and classified E.coli strains carriers Type 1 fimbriae, from patients diagnosed with childhood diarrhea, aged up to five years and in vivo, using 170 albino male Wistar rats divided into four groups: control (n = 50), Bio-Mos (n = 40), Actigen (n = 40) and Immunowall (n = 40), as an experimental model and divided in 08 studies: (1) MOS effects on the gut microbiota, through the incidence of Lactobacillus, Bifidobacterium and Escherichia coli bacteria in faeces; (2) intestinal histomorphometry evaluation; (3) determining the calcium, phosphorus, magnesium, iron, copper, zinc and selenium absorption, through the serum, liver and kidney concentrations; (4) evaluation the femur bone mineral density and bone mineral content; (5) blood biochemical evaluation processes by determining global and specific leukocyte counts, total protein, albumin, globulin, hemoglobin, urea and glucose; (6) to assess the effect on the reproductive system, by evaluating the testicles, seminal vesicles, prostate and adrenal glands cells proliferation; (7) determination testosterone and cortisol plasma concentrations; and (8) evaluation femur mineral content by using Raman spectroscopy. The experimental installation will be done in the Experimental Animal Facility at the University of Western São Paulo (UNOESTE) in a completely randomized design and will last for 63 days, 07 days for adaptation to the management and the basal diet and 56 days to treatment. The material harvest will occur in five moments: D0, D14, D28, D42 and D56, and 10 animals will be sacrificed on day 0 and every 14 days, 10 animals in each group. The harvest samples will be analyzed, in partnership, with the State University "Júlio de Mesquita Filho" (UNESP) - Campus Presidente Prudente/SP and Araçatuba/SP and with the Center for Nuclear Energy in Agriculture (CENA - USP) - Piracicaba/SP.
