Antifungal activity of phenolic enriched fraction of Buchenavia tomentosa and some isolated substances before and after encapsulation in Beta-cyclodextrin


The searching for new therapeutic alternatives for oral candidosis is of utmost importance. In this context, the aim of this study will be to evaluate the in vitro antifungal effectiveness of the phenolics enriched fraction (FE) of Buchenavia tomentosa leaves and isolates substances galic acid, corilagin, kaempferol and vitexin detected in the bioactive crude aquous extract of B. tomentosa and that will be evaluated in the FE. For the most active isolated substances and FE, the encapsulation in beta-cyclodextrin will be performed and their biological activity will be evaluated in vitro and in vivo. For this purpose, the aquous extract of the B. tomentosa leaves will be prepared and the fractions containing constituents with different polarities will be separed. Enriched fraction will be chemically analyzed by ESI-MS. This will be evaluated regarding the antifungal activity by assays of minimal inhibitory concentration and biofilm erradication using C. albicans. Then, the activity of these substances on C. albicans virulence factors will be evaluated (biofilm formation, proteinase and phospholipase production, morphogenesis, adherence to epithelial oral cells and expression of SAPs, HWP1 e ALS). Chemical, microbiological, citotoxicity and antifungal activity evaluations in vivo of the encapsulates will be performed. Data will be evaluated by descriptive statistics and analysis of normal distribution. Based on these preliminary analyses, the most suitable statistical tests will be selected.
