Bioassay-guided phytochemical study of extracts of Manilkara zapota L. and Labramia bojeri A. with activity against the Strongyloides venezuelensis demonstrated by in vitro assays

Abstract

Strongyloidiasis is a parasitic and infectious disease caused by geohelminth Strongyloides spp. It is typically located in the duodenum and proximal jejunum, it can spread to several organs is potentially fatal, especially for immunosuppressed patients. S. stercoralis has a high prevalence among humans, with about 100 million people infected worldwide. The treatments available using thiabendazole, cambendazole, albendazole and ivermectina, such drugs however are associated with high failure rates and new drugs more effective, less toxic and with less environmental impact should be investigated. Recent studies by our research group have shown promising results of the ethanol extracts of leaves of Manilkara zapota (Sapoti) and Labramia bojeri (Abricó-da-praia), fruit trees belonging to family Sapotaceae, in vitro assays against S. venezuelensis, compared to anthelmintics on the market. This project focuses on the isolation and identification of compounds responsible for anthelmintic activity of these plants against S. venezuelensis. The isolation of compounds will be bioguide by assays in vitro, for later carrying out the tests in vivo. The aqueous solubility of compounds and/or fractions will be increased from the complexation with cyclodextrins and formation of solid dispersions using polyvinylpyrrolidone (PVP) for enhance the bioavailability in vivo of the preparation. This project could lead to obtaining fractions and/or isolated compounds with potential anthelmintic activity in vitro and in vivo against S. venezuelensis from the leaves of M. zapota and/or L. bojeri (AU)
