Ecology and etiology of fibropapillomatosis in Chelonia mydas (Testudines, Cheloniidae) populations: structure of populations and the role of polychlorinated biphenyls and toxic potentially metals for the development of diseases

Abstract

The green turtle (Chelonia mydas) feeds and nests on the Brazilian coast and have been affected by fibropapillomatosis, wich is characterized of benign skin tumors and is one of the greatest threats to the survival of this species. Studies suggest virus associated with population, genetics and environmental factors. At this study, we will have access to green turtle populations from various Brazilian coast spots and (will be) determined haplotype of C. mydas populations with and without fibropapillomas from feeding areas. We will collect biometry data from green turtles, quantity and distribution of the tumors and associated with population haplotypes, CFPHV (Chelonid fibropapilloma-associated herpesvirus) detected, polychlorinated biphenyls (PCBs) and toxic potentially metals concentrations (Pb, Cd, Zn e Cu). The general aim is to elucidate the relations between C. mydas population and the role of population structure, CFPHV and environmental pollutants in the development of disease. The specifics aims are: 1- to assess the leukocytes function by flow cytometry; 2- to investigate the PCBs presence in blood samples and solid tissues by gas chromatograph; 3- to establish correlations between cell function and PCBs concentrations; 4- to investigate the metals presence and how they interfere in the sanity and development of disease; 5- to detect and to characterize the herpesvirus genic sequence associated with fibropapillomatosis (CFPHV) by moleculars methods and classic methods of virologic and hystopathologic diagnosis 7- to determine the structure of C. mydas populations from feeding areas and associate with (virus) CFPHV detected, toxic potentially metals presence (Pb, Cd, Zn e Cu) and severity and development of disease. The aims of present study enlarge the understood about the ecology and etiopathogeny of fibropapillomatosis, enabling procedures for future control of the disease in Brazilian areas and the preparation of management strategies for conservation of this species.
