Abstract
In Brazil the culture of sugarcane has expanded to areas with prolonged periods of water shortage, which has limited the production. The best way to avoid this problem is the use of stress-tolerant cultivars. This study aims the analysis of the molecular behavior of plants subjected to different periods of water limitation, and to determine the variables which could be used, independently or in combination, as an effective tool to differentiate and identify cultivars tolerant to prolonged water stress. It also expects to generate new knowledge to promote the development and competitiveness of this culture, and contribute to the formation of specialized human resources in this area. To achieve this objective, plants of two contrasting cultivars (tolerant and sensitive) will be grown in 50-liter pots in a greenhouse and subjected to three well controlled soil water potentials (assay composed by 6 treatments and 3 replications). The molecular parameters will be monitored at 30, 60 and 90 days after treatment application. After that, molecular analysis will be carried out using the technique of RNA-Seq for identifying differentially expressed genes in the cultivars under different treatments, and some of them validated by real time PCR. The results of this proposal will be important for better understanding the responses and tolerance mechanisms of these plants to prolonged water stress and help establishing secure methods to identify and recommend cultivars which are really tolerant to water limitation.
