Abstract

Schistosomiasis remains a serious public health problem in many developing countries. Reports show an increasing emergence of Schistosoma mansoni strains resistant to Praziquantel, the current drug of choice to the Schistosomiasis treatment.Recently, it was identified and characterized the apoptosis pathway mediated by Bcl-2 on Schistosoma mansoni and Schistosoma japonicum, suggesting this pathway as a new target for drugs schistosomicidal. Previous studies have shown that curcumin, which is a compound extracted from the rhizome of Curcuma longa plant has activity both in vitro and in vivo. Furthermore, studies with other organisms have shown curcumin generates reactive oxygen species (ROS) and induces apoptosis. Thus, this study aims to evaluate the induction of apoptosis in adult worms of S. mansoni. For this purpose, it will evaluate the effect of the compound on the viability of the parasites to determine the concentrations and time that will be used in the other experiments. Through microscopy, molecular and biochemical methods will be evaluated the morphology of the parasites, DNA fragmentation, the anti-apoptotic and pro-apoptotic gene expression and enzymatic activity for caspase 3 and 7 on S. mansoni adult worms after incubation with curcumin. In order to evaluate oxidative stress, some parameters will be determined as the activity of antioxidant enzymes superoxide dismutase (SOD), glutathione peroxidase (GPx) Glutathione Reductase (GR) and glutathione S transferase (GST), and will be determined the anion superoxide production and the levels of protein carbonyl. Thus, this research project will promote inter and multidisciplinary development which will be important to understand of the biology of the parasite and mechanisms by curcumin exerts it's in vitro schistosomicidal effect.
