Prospection of anti-parasitic and anti-tumor metabolites from plant species of the Atlantic Forest

Abstract

In the previous project "Principles with potential anti-Leishmania species in the Atlantic Forrest: evaluation, isolation and molecular characterization" was evaluated the therapeutic potential of medicinal plant extracts derived from the Atlantic Forest in the treatment of cutaneous and visceral leishmaniasis. Thus, we selected several plant species with potential for the discovery of anti-Leishmania substances by studying the in vitro efficacy of extracts, fractions and pure compounds obtained from plant species collected in the Atlantic Forest against promastigote and amastigote forms of Leishmania chagasi and L. amazonensis.In this project we intend to continue the isolation of substances responsible for the activity detected in extracts using bioassay-guided fractionation by testing anti-Leishmania activity, in addition to expanding the evaluation to another anti-parasitic activity against Trypanosoma cruzi in particular, and evaluate also cytotoxic activity against the various tumor cell lines. Additionally we intend to conduct specific tests to define the mode of action of substances with proven anti-parasitic activity in relation to the parasite, particularly the evaluation of membrane pore and ultrastructural studies, and for that show anti-tumor activity will be conducted specific tests to define the mechanism of action and effect on metabolic processes related to cancer development.For the isolation and molecular characterization of the active principles in the extract will perform the chromatographic fractionation, using various separation techniques followed by spectroscopic analysis to determine structural bioactive substances. Thus, this project aims to add new compounds with proven anti-parasitic activity restricted to the therapeutic arsenal, providing tools for the development of new drugs for these neglected diseases, and presents new prototypes of drugs that can be used in cancer therapy (AU)
