Abstract

Preeclampsia is the leading cause of mortality and morbidity among pregnant women in Brazil and several countries. The pathophysiology understanding is essential to decrease these numbers. Regulatory mechanisms of gene expression, in particular microRNAs may contribute to knowledge. The first objective of this project is to compare the circulating microRNAs levels using PCR-array between healthy pregnant and preeclamptic matched by the risk factors. MicroRNAs are highly stable in the circulation through of its complexing with proteins or vesicles, allowing their use as biomarkers. After PCR-array, we will validated the findings in samples from some Brazilian cities, as well as the profile of microRNAs during a healthy pregnancy and postpartum. The next step is genotype polymorphisms located in the 3'UTR (microRNA binding region) of the target gene that may influence the microRNA binding. Furthermore, the protein measurement of target gene is essential because often the contribution of this pathway is partial or total, and this should be evaluated. Aspects relating to the origin (microparticles from the placenta, exosomes released from other cell types, microRNAs complexed with proteins or current) and functionality of microRNA will also be evaluated. With respect to the latter, we will incubate plasma of healthy pregnant or preeclamptic women, and molecules that mimic or antagonize microRNA with endothelial cells (HUVECs) and see how this process modifies the gene expression of important pathways in the preeclampsia pathophysiology. We believe that the data obtained in this project will provide relevant information to the study of preeclampsia in the global context. (AU)
