Bone repair evaluation of critical rats calvarial deffects filled with Bio-Oss or Bio-Oss collagen

Abstract

A limiting factor for dental implants placement is the lack of bone in the alveolar ridges and usually occurs in patients who have suffered dentoalveolar trauma, traumatic dental extractions, dental congenital absence, or other pathologies involving the maxilla and mandible. The inappropriate bone volume undertakes primary stability. Extensive bone defects may require a large autogenous bone tissue to fill out, increasing the morbidity of the procedure. Currently, the trend is to develop and use biomaterials that accelerate or at least allow normal and complete the bone repair defects, reducing the failure rate postoperatively. Bio-Oss ® is a bone substitute obtained from mineral part of bovine bone which favors proliferation of blood vessels and bone cells migrating through the system in its wide mesh of interconnecting pores, and may or may not have collagen to improve its osteoconductive properties. Therefore, the aim of this project will be to assess the osteoconductive potential of lyophilized bovine bone (Bio-Oss ®) and lyophilized bone plus 10% collagen (Bio-Oss Collagen ®) in the repair bone defect of rat calvaria, at 7, 30 and 60 days, through histometric analysis. For this purpose, 30 adult male rats will be divided into two groups (n = 5 ) being euthanized at three times of the experiment (7, 30 and 60 days) after surgery. In each animal, a critical defect in the calvaria (7 mm) will be perfomed with a trephine, and the Group Bio-Oss (n = 15), wherein the critical bone defect will be filled with Bio-Oss and Group Bio-Oss Collagen (n = 15), will be filled with lyophilized bone plus collagen. The central area of bone defect will be evaluated. The obtained data will be converted into absolute values, from pixels to percentage values, to minimize the interference of the size difference of negative slides stained with hematoxylin and eosin (HE). To compare the mean values obtained from the different groups and experimental periods, ANOVA test will be applied. Significance level of 5% (p <0.05) will be adopted. (AU)
