Abstract
Currently asthma and Chronic Obstructive Pulmonary Disease (COPD) have a high prevalence and a high economic and social cost. In this context, several experimental models have been proposed, aiming to advance knowledge about the pathophysiological processes of asthma and COPD, as well as the discovery of new therapeutic options. Therapy with Low Power Laser (LBP) is a relatively new effective therapy, with very low cost and no side effects. In this project we aim to study some parameters in animals with asthma and COPD undergoing therapy with LBP. The protocol used for the induction of asthma will be administered OVA subcutaneously (days 0 and 14) and intranasal OVA (three times / day for five weeks) or HDM intranasally (on days 0, 7, 14, 21, 28, 35 and 42). On days 43 (OVA) and 50 (HDM), animals will be sacrificed for morphologic and functional analysis of the lung. The protocol used for the induction of COPD is inhaling C57BL/6 mice for 60 days (2 times / day) with cigarette smoke. On day 61, animals will be sacrificed for morphologic and functional analysis of the lung. In both models, we will study functional and structural parameters, such as the release of cytokines and chemokines (IL-4, IL-5, IL-10, IL-13, MCP-1, TNF-±, IL-1², IL-6 and IL-10. We will also evaluate the participation of the transcription factors STAT6, GATA3 and STAT4 and NF-ºB (intact and phosphorylated), MMP-9 and MMP-12, TIMP-1 and TIMP-2 and the evaluation lung mechanics and airway hyperresponsiveness in both experimental models will be performed.
