Transcriptome characterization and SNPs discovery in species of Serrasalmidae: genetic subsidies for aquaculture

Abstract

Species of Serrasalmidae, pacu Piaractus mesopotamicus, pirapitinga Piaractus brachypomus and tambaqui Colossoma macropomum, represent the major group of native fish intended for breeding programs, due to their market acceptance (national and international) and because they are the main fish produced currently. However, the genetic information available for these species is insufficient. Transcriptome characterization using next-generation sequencing (NGS) technologies is an efficient resource (fast and low cost) to generate high volume of genetic information and to prospect SNPs (single-nucleotide polymorphisms) markers. The purpose of this project is to perform the transcriptome sequencing and characterize SNPs markers (intra and interspecific) in pacu, pirapitinga and tambaqui, which will be made by NGS. The construction of cDNA libraries will be obtained from the total RNA extracted of liver of 10 individuals for each species. The transcriptome sequencing will be held at the Roche/454 platform. Bioinformatics analyzes will be made to de novo assembly, functional annotation, discovery and classification of SNPs. The interspecific SNPs will be validated through Multiplex-PCR method. In addition to the introduction of new genetic technologies to the Brazilian aquaculture industry, the expect result is that these informations of transcriptome and SNPs might be used for the construction of genetic maps combined with studies of animal performance, which will accelerate the genetic improvement and increase the productivity of this important group of fish.
